Plasmid restriction fragment analysis

Restriction enzyme analysis may be used to evaluate genetic stability of a plasmid construct by evaluating the restriction profile in comparison to a reference or theoretical profile.


The plasmid will be extracted from test article cells and digested with restriction enzymes of up to three enzymes. The digests and an uncut plasmid control will be electrophoresed along with the standard molecular weight marker. After the gel is stained and imaged, the restriction fragments will be analyzed and the observed fragment sizes will be reported and compared to the calculated fragment sizes from the plasmid reference sequence for confirmation of construct integrity.
